RESUMEN

Vibrio parahaemolyticus is a gram-negative facultative anaerobic bacterium implicated as the causative agent of several shrimp diseases. As part of the effort to provide biocontrol and cost-effective treatments, this research was designed to elucidate the effect of Morinda citrifolia fruit extract on the immunity of Penaeus vannamei postlarvae (PL) to V. parahaemolyticus. The methanol extract of M. citrifolia was vacuum evaporated, and the bioactive compounds were detected using gas chromatography‒mass spectrometry (GC‒MS). Thereafter, P. vannamei PL diets were supplemented with M. citrifolia at different concentrations (0, 10, 20, 30, 40, and 50 mg/g) and administered for 30 days before 24 h of exposure to the bacterium V. parahaemolyticus. A total of 45 bioactive compounds were detected in the methanol extract of M. citrifolia, with cyclononasiloxane and octadecamethyl being the most abundant. The survival of P. vannamei PLs fed the extract supplement was better than that of the control group (7.1-26.7% survival greater than that of the control group) following V. parahaemolyticus infection. Shrimp fed 50 mg/g M. citrifolia had the highest recorded survival. The activities of digestive and antioxidant enzymes as well as hepatopancreatic cells were significantly reduced, except for those of lipase and hepatopancreatic E-cells, which increased following challenge with V. parahaemolyticus. Histological assessment of the hepatopancreas cells revealed reduced cell degeneration following the administration of the plant extracts (expecially those fed 50 mg/g M. citrifolia) compared to that in the control group. Therefore, the enhanced immunity against V. parahaemolyticus infection in P. vannamei could be associated with the improved hepatopancreas health associated with M. citrifolia fruit extract supplementation.

Marine periphytic ciliates play a pivotal role in shaping coastal ecosystems dynamics, thereby acting as robust biological indicators of aquatic ecosystem health and functionality. However, the understanding of the effects of veterinary antibiotics on composition and structure of periphytic ciliate communities remains limited. Therefore, this research investigates the influence of the veterinary antibiotic nitrofurazone on the community dynamics of marine periphytic ciliates through bioassay experiments conducted over a one-year cycle. Various concentrations of nitrofurazone were administered to the tested ciliate assemblages, and subsequent changes in community composition, abundance, and diversity were quantitatively analyzed. The research revealed significant alterations in periphytic ciliate communities following exposure to nitrofurazone. Concentration-dependent (0-8 mg L-1) decrease in ciliates abundance, accompanied by shifts in species composition, community structure, and community patterns were observed. Comprehensive assessment of diversity metrics indicated significant changes in species richness and evenness in the presence of nitrofurazone, potentially disrupting the stability of ciliate communities. Furthermore, nitrofurazone significantly influenced the community structure of ciliates in all seasons (winter: R2 = 0.489; spring: R2 = 0.666; summer: R2 = 0.700, autumn: R2 = 0.450), with high toxic potential in treatments 4 and 8 mg L-1. Differential abundances of ciliates varied across seasons and nitrofurazone treatments, some orders like Pleurostomatida were consistently affected, while others (i.e., Strombidida and Philasterida) showed irregular distributions or were evenly affected (e.g., Urostylida and Synhymeniida). Retrieved contrasting patterns between nitrofurazone and community responses underscore the broad response repertoire exhibited by ciliates to antibiotic exposure, suggesting potential cascading effects on associated ecological processes in the periphyton community. These findings significantly enhance the understanding of the ecological impacts of nitrofurazone on marine periphytic ciliate communities, emphasizing the imperative for vigilant monitoring and regulation of veterinary antibiotics to protect marine ecosystem health and biodiversity. Further research is required to explore the long-term effects of nitrofurazone exposure and evaluate potential strategies to reduce the ecological repercussions of antibiotics in aquatic environments, with a particular focus on nitrofurazone.

Polyalthia longifolia var. angustifolia Thw. (Annonaceae), is a famous traditional medicinal plant in Asia. Ample data specifies that the medicinal plant P. longifolia has anticancer activity; however, the detailed mechanisms of action still need to be well studied. Recent studies have revealed the cytotoxicity potential of P. longifolia leaf against HeLa cells. Therefore, the current study was conducted to examine the regulation of miRNAs in HeLa cancer cells treated with the standardized P. longifolia methanolic leaf extract (PLME). The regulation of miRNAs in HeLa cancer cells treated with the standardized PLME extract was studied through Illumina, Hi-Seq. 2000 platform of Next-Generation Sequencing (NGS) and various in silico bioinformatics tools. The PLME treatment regulated a subset of miRNAs in HeLa cells. Interestingly, the PLME treatment against HeLa cancer cells identified 10 upregulated and 43 downregulated (p < 0.05) miRNAs associated with apoptosis induction. Gene ontology (GO) term analysis indicated that PLME induces cell death in HeLa cells by inducing the pro-apoptotic genes. Moreover, the downregulated oncomiRs modulated by PLME treatment in HeLa cells were identified, targeting apoptosis-related genes through gene ontology and pathway analysis. The LC-ESI-MS/MS analysis identified the presence of Vidarabine and Anandamide compounds that were previously reported to exhibit anticancer activity. The findings of this study obviously linked the cell cytotoxicity effect of PLME treatment against the HeLa cells with regulating various miRNAs expression related to apoptosis induction in the HeLa cells. PLME treatment induced apoptotic HeLa cell death mechanism by regulating multiple miRNAs. The identified miRNAs regulated by PLME may provide further insight into the mechanisms that play a critical role in cervical cancer, as well as novel ideas regarding gene therapeutic strategies.

This study focuses on the relationship between myostatin (MyoS), myogenin (MyoG), and the growth hormone/insulin-like growth factor-1 (GH/IGF-1) axis for muscle growth and histopathological changes in muscle after an Aeromonas hydrophila infection. A total number of 90 Nile tilapia (55.85 g) were randomly allocated into two equal groups of three replicates each. The first group was an uninfected control group that was injected intraperitoneally (ip) with 0.2 ml phosphate buffer saline (PBS), while the second group was injected ip with 0.2 ml (1.3 × 108 CFU/ml) Aeromonas hydrophila culture suspension. Sections of white muscle and liver tissues were taken from each group 24 h, 48 h, 72 h, and 1 week after infection for molecular analysis and histopathological examination. The results revealed that with time progression, the severity of muscle lesions increased from edema between bundles and mononuclear inflammatory cell infiltration 24 h post-challenge to severe atrophy of muscle bundles with irregular and curved fibers with hyalinosis of the fibers 1 week postinfection. The molecular analysis showed that bacterial infection was able to induce the muscle expression levels of GH with reduced ILGF-1, MyoS, and MyoG at 24 h postinfection. However, time progression postinfection reversed these findings through elevated muscle expression levels of MyoS with regressed expression levels of muscle GH, ILGF-1, and MyoG. There have been no previous reports on the molecular expression analysis of the aforementioned genes and muscle histopathological changes in Nile tilapia following acute Aeromonas hydrophila infection. Our findings, collectively, revealed that the up-and down-regulation of the myostatin signaling is likely to be involved in the postinfection-induced muscle wasting through the negative regulation of genes involved in muscle growth, such as GH, ILGF-1, and myogenin, in response to acute Aeromonas hydrophila infection in Nile tilapia, Oreochromis niloticus.

Mud crabs (genus Scylla) are gaining attention as potential aquaculture species due to their lucrative market price and high demand. One of the essential components of mud crab culture is broodstock quality. The optimisation of mud crab broodstock culture currently focused on its nutritional aspects and common rearing parameters, including stocking density and temperature. The use of sandy substrate to induce egg extrusion in female Scylla broodstock is common; however, its optimisation has never been conducted. This study investigated (1) the substrate association of two Scylla species (S. olivacea, S. paramamosain) during broodstock conditioning until egg extrusion when the choices of fine (66.99 ± 14.48 µm) and coarse (656.17 ± 38.13 µm) sands were given; (2) the female reproductive output when S. olivacea females were individually exposed to either fine sand, coarse sand, or no sand treatments. Females, spawners and non-spawners, of S. olivacea and S. paramamosain were associated with fine sand and none was observed to bury in the coarse sand tray. The occurrence of egg extrusion was not significantly different between species but moderately associated with the duration of visits (stayed in sand for 1 d, 2 d, ≥3 d). The final incubation period in the sand tray was more than 2 days in all cases, except for one female S. paramamosain (buried in the sand for 1 day prior to egg extrusion). When no choice was available, the highest percentage (58.3%) of females extruded eggs in fine sand treatment, followed by coarse sand treatment (33.3%), and no sand treatment (8.3%). Sand type influenced the weight of egg clutch, total egg number, fecundity, and clutch size. These results suggest that fine sand (<70 µm) substrate should be incorporated into Scylla broodstock rearing to maximise female reproductive output.

Macroinvertebrate community in the intertidal setup plays an important role in coastal ecosystem functions and biogeochemical cycle. However, different land use pattern may influence on their community structure, diversity, and composition in the coastal ecosystems. Using Van-Veen grab sampler, 60 sediment samples were seasonally collected from mangroves-dominated, aquaculture-dominated, and anthropogenically affected area in the lower intertidal zone of the Kohelia channel of Bangladesh, the Northern Bay of Bengal. We have tasted the variation in sediment properties across three land-use types in this intertidal habitat. To understand the patterns of benthic macroinvertebrate distribution, a neutral community model was applied. Our results showed that community composition and biodiversity of the benthic macroinvertebrate communities varied significantly between mangrove-dominated area with anthropogenically affected areas among the four seasons. The neutral community model revealed that community assembly of benthic macroinvertebrates in the lower intertidal habitats is structured by stochastic processes while sediment properties have significant influence on species distribution and interactions. Results suggested that land-use changes altered sediment properties and could change the diversity and distribution of the macroinvertebrate communities in the lower intertidal habitats.

The objective of the present study was to evaluate the growth performance and genetic variation in diallel crosses of Ariza labeo (Labeo ariza) originating from three geographically separated rivers (Atrai, Jamuna and Kangsha) in Bangladesh. Intra (G1K♀K♂, G2J♀J♂, and G3A♀A♂) and inter (G4K♀A♂, G5K♀J♂, G6A♀K♂, G7A♀J♂, G8J♀K♂, and G9J♀A♂) stocks were produced following diallel cross (sex ratio-1:1 and n = 48; 16 from each river). Reproductive and growth performance, muscle cellularity and genetic variation following genotyping of eight microsatellite markers (Lr1, Lr2, Lr3, Lr22, Lr24, Lr27, Lr28 and Lr29) and analysis of all crossbreeds was performed. The fertilization (95% ± 2.11%), hatching (88% ± 1.03%), and survival rates (82% ± 1.88%) of G4K♀A♂ were higher compared to other groups. With respect to length and weight gains (2.67 ± 0.4 cm and 3.39 ± 0.2 g), SGR (3.23% ± 0.20%), and heterosis (8.87% and 24.74%) G4K♀A♂ was the superior group. A higher number of hyperplastic muscle fibers, mean number of alleles (2.75) and mean observed heterozygosity (0.417) from G4K♀A♂ could be interpreted to mean that G4K♀A♂ comprise better performance efficiency compared to others and are considered for continuing the L. ariza stock improvement program.

Okra (Abelmoschus esculentus L. (Moench) plays a significant role in humans nutrition because its fresh leaves, stems, flowers, pods and seeds, are used for multiple purposes. The present study attempted to determine the spatial variations in biochemical attributes of osmoprotectants and the oxidative defense system of okra plants. Samples of soil and okra plants (leaves and fruits) were collected from three different locations: Faisalabad region-1 (7 JB-I), Faisalabad region-2 (7 JB-II) and Pindi Bhattian. Chlorophyll contents, glycine betaine (GB), ascorbic acid (AsA), total phenolics, hydrogen peroxide (H2O2), proline, and malondialdehyde (MDA) contents were analyzed in the leaves and fruits of okra plants. Soil analyses showed that pH, electrical conductivity (EC), phosphorus (P), potassium (K), iron (Fe), and saturation of soil were higher in Faisalabad region 2, while organic matter, sand, Zn, and Cu were higher in the Pindi Bhattian region. The results from okra leaves showed that Pindi Bhattian had higher chlorophyll a, GB and H2O2 contents, while Faisalabad region 1 had a higher ratio of chlorophyll a/b compared to the other regions. However, Faisalabad regions 2 and 1 had higher leaf phenolic contents, Faisalabad regions 1 and 2 showed higher leaf proline contents, and Faisalabad region 2 possessed higher AsA and MDA contents. Analyses of okra fruits showed that Faisalabad region 2 had higher chlorophyll a and total chlorophyll contents, while Faisalabad region 1 had higher chlorophyll b contents. Faisalabad region 2 and Pindi Bhattian had higher ratios of chlorophyll a/b, and Faisalabad region 1 showed higher phenolic, AsA, H2O2, and MDA contents of okra fruit, whereas the Faisalabad regions exhibited higher proline and GB contents than the Pindi Bhattian region. Overall, okra leaves and fruits showed better responses in the Faisalabad regions, and these results may be used to screen for okra cultivars with better tolerance under different environmental conditions.

Environmental contamination of chromium (Cr) has gained substantial consideration worldwide because of its high levels in the water and soil. A pot experiment using oil seed crop (rapeseed (Brassica napus L.)) grown under different levels of tannery wastewater (0, 33, 66 and 100%) in the soil using the foliar application of zinc (Zn) and iron (Fe)-lysine (lys) has been conducted. Results revealed that a considerable decline in the plant growth and biomass elevates with the addition of concentrations of tannery wastewater. Maximum decline in plant height, number of leaves, root length, fresh and dry biomass of root and leaves were recorded at the maximum level of tannery wastewater application (100%) compared to the plants grown without the addition of tannery wastewater (0%) in the soil. Similarly, contents of carotenoid and chlorophyll, gas exchange parameters and activities of various antioxidants (superoxidase dismutase (SOD), peroxidase (POD), catalase (CAT) and ascorbate peroxidase (APX)) were also reduced significantly (P < 0.05) with the increasing concentration of tannery wastewater (33, 66 and 100%) in the soil. In addition, a combined application of Zn and Fe-lys reduced the accumulation and uptake of toxic Cr, while boosting the uptake of essential micronutrients such as Zn and Fe in different tissues of the plants. Results concluded that exogenous application of micronutrients chelated with amino acid successfully mitigate Cr stress in B. napus. Under field conditions, supplementation with these micronutrient-chelated amino acids may be an effective method for alleviating metal stress in other essential seed crops.

Rapid urbanization with an increasing rate of urban built-up area is decreasing urban green space resulting in changing urban microclimate conditions showing increasing land surface temperature. A better understanding of these effects is important to formulate effective strategies in addressing the impact of increasing built-up area. Land surface temperature patterns in an urbanized city in Bangladesh (Mymensingh district) were investigated using Landsat satellite sensor data from 1988 to 2016. A total of nineteen Landsat satellite images were used to retrieve land surface temperature (LST), normalized difference vegetation index (NDVI), and normalized difference built-up index (NDBI). The radiative transfer equation (RTE) model was applied to derive LST for the years 1988, 1992, 1999, 2004, 2008, 2012, and 2016. Further, the Landsat-derived LST results were compared with MODIS Terra satellite outputs (MOD11A1) for the validation of our study results. Our results showed NDVI higher in 2008 and lower in 2004, LST maximum in 1988 and minimum in 2008, and NDBI higher in 2004 and lower in 2012. Seasonally, summer was characterized by higher LST and winter by lower LST, while NDVI was higher in autumn and lower in winter, however, NDBI was higher in winter and lower in autumn. Spatially, a relatively higher LST and NDBI was observed in the southwest, followed by central, and northern regions, whereas the trend was opposite for NDVI. Using Pearson's correlation, results showed a strong significant negative correlation between LST and NDVI and a positive significant correlation between LST and NDBI. Further, simple linear regression analysis revealed that LST decreased with increasing NDVI most quickly in 2012, followed by the years 2016, 2008, 1992, 1988, 1999, and 2004. On the other hand, LST increased with increasing NDBI most quickly in 1999, followed by the years 2016, 1988, 1992, 2012, 2004, and 2008. Thus, long-term observation suggested that urbanization had driven a decrease in green space while simultaneously increasing the land surface temperature within an urbanized area. This study has concluded that the protection of urban green spaces is needed as an effective step toward addressing adverse effects of regional climate change and desertification.

Chlorpyrifos (CP) is a pesticide used extensively in agricultural crops. Residual CP has been found in a variety of soils, vegetables and fruits indicating a serious danger to humans. Therefore, it is necessary to restrict its entry into agricultural products for food safety. A wire-house pot experiment was conducted with maize plants in biochar- and compost-amended soil (at 0.25% and 0.50%, respectively, in weight-by-weight composition) contaminated with 100 and 200 mg kg-1 of CP, respectively. Results indicated toxicity at both CP levels (with 84% growth reduction) at CP 200 mg kg-1. However, application of compost and biochar at the 0.50% level improved the fresh weight (2.8- and 4-fold, respectively). Stimulated superoxide dismutase (SOD) and peroxidase (POX) activities and depressed catalase (CAT) activity were recorded in response to CP contamination and were significantly recovered by the amendments. Both amendments significantly decreased the CP phytoavailability. With biochar, 91% and 76% reduction in the CP concentration in maize shoots and with compost 72% and 68% reduction was recorded, at a 0.50% level in 100 and 200 mg kg-1 contaminated treatments respectively. Compost accelerated the CP degradation in postharvest soil. Therefore, biochar and compost amendments can effectively be used to decrease CP entry in agricultural produce by reducing its phytoavailability.

Ammonia is a critical hazardous nitrogen metabolic product in aquaculture. Despite trials for its control, ammonia intoxication remains one of the most critical issues to overcome. In this study, we explored the modulatory effect and potential mechanism by which Yucca schidigera extract (YSE) can ameliorate ammonia intoxication-induced adverse effects on tilapia health and metabolism. A total number of 120 Nile tilapia were evenly assigned into four groups with three replicates each. The first group served as normal control group; the second group was exposed to ammonia alone from the beginning of the experiment and for four weeks. The third group was supplied with YSE in water at a dose of 8 mg/L and exposed to ammonia. The fourth group was supplied with YSE only in water at a dose of 8 mg/L. YSE supplementation succeeded in improving water quality by reducing pH and ammonia levels. Moreover, YSE supplementation markedly alleviated chronic ammonia-induced adverse impacts on fish growth by increasing the final body weight (FBW), specific growth rate (SGR), feed intake and protein efficiency ratio (PER) while reducing the feed conversion ratio (FCR) via improvements in food intake, elevation of hepatic insulin-like growth factor (ILGF-1) and suppression of myostatin (MSTN) expression levels with the restoration of lipid reserves and the activation of lipogenic potential in adipose tissue as demonstrated by changes in the circulating metabolite levels. In addition, the levels of hepato-renal injury biomarkers were restored, hepatic lipid peroxidation was inhibited and the levels of hepatic antioxidant biomarkers were enhanced. Therefore, the current study suggests that YSE supplementation exerted an ameliorative role against chronic ammonia-induced oxidative stress and toxic effects due to its free radical-scavenging potential, potent antioxidant activities and anti-inflammatory effects.

The increasing market demand for decapods has led to a considerable interest in cultivating decapod species at a larger scale. Following the development of hatchery technologies, most research has focused on the development of formulated feeds for commercially farmed decapods once they enter the juvenile stages. The use of formulated feed for decapods at a commercial scale is still in the early stages. This is probably because of the unique feeding behavior that decapods possess: being robust, slow feeders and bottom dwellers, their feeding preferences change during the transition from pelagic larvae to benthic juveniles as their digestive systems develop and become more complex. The current practice of decapod aquaculture involves the provision of juveniles with food such as natural diet, live feed, and formulated feed. Knowledge of nutrient requirements enables diets to be better formulated. By manipulating the levels of proteins and lipids, a formulated feed can be expected to lead to optimal growth in decapods. At the same time, the pellet's physical characteristics are important factors to be considered upon formulating commercially farmed decapod feeds, considering the unique feeding behavior of the decapod. However, most published studies on decapod nutrition lack data on the physical characteristics of the feed types. Thus, it is difficult to establish a standard feed formulation that focuses on the physical pellet properties. Moreover, careful consideration must be given to the feeding behavior of species, as decapods are known as bottom feeders and are robust in terms of handling feed. Information on the pellet forms, diet composition, and unique feeding behaviors in commercially farmed decapods is gathered to suggest potential better formulated diets that can optimize growth and reproduction. Thus, the purpose of this review is to summarize the information that has been published to date and to come up with suggestions on ways to improve the feed formulation in decapods that comply with their feeding behavior and nutrient requirements. Further research is needed to explore the potential of the pelleted feed at the adult stage so the decapod can take full advantage of the nutrients present in the pellets.

RESUMEN

Seagrass habitats are considered to be some of the most biodiverse ecosystems on the planet and safeguard some ecologically and economically important fauna, amongst which are some globally threatened species, including dugong. Malaysian seagrass ecosystems are not widespread, but their existence supports some significant marine fauna. A rigorous zooplankton study was conducted from May 2016 to February 2017, in the seagrass habitat of Lawas, Sarawak, Malaysia, to examine their temporal composition and diversity, together with their ecological influences. A total of 45 zooplankton species from 13 significant groups were recorded in the seagrass habitat. The population density of zooplankton ranged between 2,482 ind/m³ and 22,670 ind/m³ over three different seasons. A single zooplankton copepod was found to be dominant (47.40%), while bivalves were the second largest (31.8%) group in terms of total abundance. It was also noticed that the average relative abundance (0.62) and important species index (62.08) of copepods were higher than for other groups that exist in the seagrass meadow, whereas copepod Parvocalanus crassirostris showed both the highest average relative abundance (0.41) and the highest important species index (41.15). The diversity (H') and richness index of the intermediate season were found to be highest due to favourable physico-chemical conditions. Within the referred seasonal cluster, the wet and dry seasons were almost similar in terms of species abundance, while the intermediate season was distinct, with high species diversity backed by ANOSIM analysis results. Copepod and bivalves formed one group with a common similarity level of 0.80. The CCA (Canonical Correspondence Analysis) model established that abiotic factors, especially turbidity, NO2, rainfall, dissolved oxygen and pH were significantly correlated with abundance of individual groups of zooplankton. Zooplankton assemblage and abundance in Lawas were found to be very rich in multiple seasons, indicating that the productivity of uninterrupted seagrass habitat might be high and the system rich in biodiversity.
